More than 20 firefighters pulled two children and two adults from a blaze at a converted block of flats in Leytonstone.
The property, a house converted into several flats in Birkbeck Road, caught alight at around 3am on Thursday morning.
One man and one woman had been able to escape the house before the arrival of London Fire Brigade and were treated at the scene by London Ambulance Service.
One man, one woman and two children were led to safety by firefighters wearing breathing apparatus.
Part of a flat on the ground floor of the building was damaged by fire.
Crews from Leytonstone and Leyton fire stations were able to bring the blaze under control by 5.33am.
The cause of the fire is under investigation.
